The Honda CR-V emergency rapid response vehicle collided with a Suzuki this morning at about 9am in Mauchline Road.
The female driver of the car was rushed to hospital by ambulance after suffering pelvic injuries.
Fire and Rescue Service Ayr station manager Derek Davidson said: “The paramedic was trapped in his vehicle and suffered from back and neck pain plus an injured thumb. Fire crews assisted ambulance personnel to remove their colleague and have him taken to hospital for further assessment and treatment."
A Police Scotland spokeswoman said the pair were in "stable condition" in hospital and the injuries were not life threatening.
Mr Davidson added: “I would urge drivers to be vigilant on the road and act appropriately when you see an emergency service vehicle in the vicinity. Pull over, give a clear indication of your actions and make way in a safe and controlled manner.”
